    Title: 甘薯葉蛋白之萃取與乙醇萃取物胰島素阻抗活性之探討

    Abstract: 台灣環境條件非常適合甘藷生長,其生產潛力也較其他主要的熱帶糧食作物高,所以甘藷種植 非常普遍,被視為本土型之重要糧食作物。主持人之前曾執行農委會 70%乙醇甘藷葉萃取物抗氧化 性之探討,亦獲致相當之成果。然而,執行過程中發現,若僅只為抽取少量的萃取物而將甘藷中諸 多好的成份如蛋白質、膳食纖維棄置,殊屬可惜。若能將其回收利用,則可增加收益並達到減廢之 功能。 因此本計畫第一年將針對甘藷葉蛋白萃取之條件及其物理化學特性進行探討。實驗利用改變萃 取溫度、萃取溶液 pH 值、萃取時間與萃取體積重量比例四項因子進行探討。並利用反應曲面法探 討最適萃取條件。所得甘藷葉蛋白並分別進行各項物化功能測試。第二年進行甘藷葉 70%乙醇萃取 物之生理功能性探討,主要以改善葡萄糖之攝入作為實驗探討之對象。實驗利用小鼠肝臟上皮細胞 FL83B 為細胞株,先進行細胞存活率試驗 (MTT assay),接著進行薯葉乙醇萃物對具胰島素阻抗性 FL83B 細胞之葡萄糖攝入試驗 (Glucose uptake),並利用西方轉印分析(Western blot)了解可能作用之 途徑。另外藉由分析總酚、類黃酮類與花青素了解其可能有效之生理功能物質來源。實驗完成期望 可增加甘藷之加工利用與附加價值。
    The environmental condition of Taiwan is suitable for sweet potato growth and the potential of economic of sweet potato is also higher than other crops. Our laboratory had been studied the antioxidative activity of 70% alcohol extract of sweet potato several years ago which was support by Council of Agriculture’s fund and had been found some variety of sweet potato own high antioxidative activity. However, during the studied time, we found after the pulp was extracted, most of the valuable composition of sweet potato such as protein, dietary fiber had been discarded. If we recycled some composition, it was going to be reduced the waste. This research will be continued to two years. The first year are going to study the extract conditions of sweet potato protein and their physicochemical property. The experiment will use extract temperature, pH of extract solution, extract time and ratio of sweet potato weight and solution as four factors to find out the extract condition of sweet potato leaf protein. Meanwhile, the response surface methodology will be used to find out the optimum extract condition. The extracted protein will investigate their physicochemical property also. The second year will going to investigate the physiological property, mainly in improve glucose uptake model, of 70% alcohol extract of sweet potato leaf. The experiment will use the FL83B cell as cell line to test the cell viability and glucose uptake in the insulin resistance cell model by 70% alcohol extract of sweet potato leaf. Then, use the Western blot method to find out the possible mechanism. Meanwhile, the total content of phenolic compounds, flavonoids and anthocyanins will be analysed to find out the possible source of glucose uptake improvement. We hope after the study finished, it will increased the economic value of sweet potato product.
