    题名: 異黃酮前趨物(DOB-3-A)在皮膚細胞中抗紫外線傷害保護作用之研究

    摘要: Deoxybenzoins (DOBs)是合成異黃酮結構化合物的前趨物,具有類似異黃酮的結構,在生物實驗和分子模型的研究中指出,DOBs可被考慮作為乳腺癌抗癌藥劑和荷爾蒙替代療法上的取代物。DOBs被證明是具有強力的抗氧化和抗發炎作用,而且比維生素E和維他命C更具有良好自由基清除能力。然而,目前較少研究在探討Deoxybenzoins結構化合物DOB-3-A對抗紫外線A的保護效果。因此,我們將利用纖維母細胞(fibroblast),探討DOB-3-A 對抗紫外線A的保護效果。經結果顯示,fibroblast在DOB-3-A前處理後,可對抗紫外線的傷害,提高細胞的存活率。而其作用機轉中,發現DOB-3-A在紫外線所活化的分裂原活化蛋白激酶表現中,具有抑制其磷酸化作用,例如: ERK、p38、JNK。此外DOB-3-A也會減少fibroblast在紫外線刺激後所產生的第二型環氧化酶(COX-2)的表現。此外,DOB-3-A會增加fibroblast在紫外線刺激後激活PI3K/AKT的訊號路徑的表現,並導致細胞不正常的持續增生。另外,fibroblast在紫外線A的刺激後,會產生H2O2,而低濃度DOB-3-A在短時間可減少H2O2產生量,而隨著DOB-3-A濃度的提高,可減少的H2O2也愈多。綜合上述結果,DOB-3-A可預防紫外線A對皮膚所造成的傷害,並有助於改善紫外線在皮膚上造成的損傷。
    Deoxybenzoins (DOBs) are the precursors of synthetic isoflavone compounds, which structure is similar to isoflavones. The researches of the biological experiments and molecule-models suggest that we can consider DOBs to replace the pharmacy of anti-breast-cancer and hormone replacement treatment. DOBs have been proved to have the useful and powerful anti-oxidation and anti-inflammation functions, and the ability to clear free radicals is even better than vitamin E and vitamin C. However, there are few researches probing into the Deoxybenzoin structural compound; that is, DOB-3-A’s protective function to resist the UVA. As a result, we utilized the fibroblast to explore DOB-3-A’s anti-UVA effects. The results showed that the fibroblast pre-treated by DOB-3-A could resist the UV damage and raise the cell survival rate. We found that DOB-3-A in UV activated mitogen activation protein kinase could inhibit the phosphorylation activity like ERK, p38 and JNK. Besides, DOB-3-A could reduce COX-2 expression gendered by the UV stimulated fibroblast. DOB-3-A could increase the PI3K/AKT signal pathway activated by the UV stimulated fibroblast and thus lead to continuously abnormal cell proliferation. Furthermore, numerous studies have demonstrated UVA induced intracellular H2O2 production in fibroblast. In our study, DOB-3-A reduced H2O2 generation after UVA exposure as dose dependence. To sum up, from the results listed above, we conclude that DOB-3-A can protect the skin against UV damage and help the skin to recover from UVA damage.
