    Abstract: 近年來,有越來越多的研究人員使用嗜甲基酵母菌Pichia pastoris當作宿主細胞來表現異源重組蛋白質。與動物細胞相比,P. pastoris具有分子基因操作容易,且易於發酵培養而達到高細胞濃度等優點。它亦是真核細胞,具有蛋白質轉譯後修飾的能力,能產生正確摺疊具有生物活性的蛋白質。而外來基因可以利用基因同源重組作用,將基因插入P. pastoris染色體而成為穩定的細胞株。此外,P. pastoris擁有強的AOX與GAP啟動子可以大量的表現異源重組蛋白質。本文將介紹此一深具潛力的P. pastoris蛋白質表現系統。

    In recent years, there has been an increasing focus on the methylotrophic yeast, Pichia pastoris, a host cell for the recombinant proteins expression. P. pastoris is easier to genetically manipulate and culture than mammalian cells, and high cell density cultivations can be easily achieved in the flasks or in the fermentors. P. pastoris is also a eukaryote, and thereby providesthe potential for producing soluble, correctly folded recombinant proteins that have undergone allthe post-translational modifications. The linearized foreign DNA can be inserted into its chromosomes via homologous recombination to generate stable cell lines. Furthermore, P. pastoris has strong AOX and GAP promoters to drive the recombinant expressions, therefore, production of high level target protein can be easily achieved. This article is to introduce the P. pastoris protein expression system.
